Macroscopic Examination (Normal Constituents)
Volume – 4-5ml
Viscosity – high
Color – cloudy grey white
pH – 7.2-8.9
Microscopic Examination
Procedure is done after liquefaction, i.e. 15-30 minutes
Motility place a drop of liquefied semen on the glass slide and cover with a cover slip. Normally 80% sperms are motile.
<50% motility of sperms- significant
Sperm count- draw semen upto 0.5mark in WBC pipette and diluting fluid upto 11 mark (dilution is 1:20). Mix well.
Diluting fluid- components:
Sodium bicarbonate - 5gms
Phenol – 1ml
Distilled water – 100 ml
Charge the counting chamber (Neubauer) and count the cells in four corner squares.
Sperm count/ml = N x 10 x 20 x 1000 / 4
Normal sperm count = 60 to 150 million / ml
Oligozoospermia = <60million / ml
