CSF is formed from the choroid plexus
Technique Lumbar puncture
Site L3L4(adults)
L4L5(children)
Procedure: CSF is often examined in the counting chamber (Neubauer’s chamber) without addition of any diluting fluid. The number of cells are counted in all the 9 large squares. Cell count should be done within 30 min of collection of specimens.
Calculation
No: of cells counted = N
Dilution = 1:1
Depth = 1/10 mm
Area = 9mm2
Cells/liter(mm) = n x 1 x 10 /9
= 1.11 x N
